As in the first session, Hector Barbera took the best time in the free practice of the 250 class in Brno, recording the reference time of 2'02.416 aboard his Pepe World Team Aprilia. The Spanish driver preceded his compatriot Alvaro Bautista and Hiroshi Aoyama, separated by just over a tenth, with Mike Di Meglio in fourth position.

Good result for Roberto Locatelli, fifth and also managing to take the lead, best Italian ahead of Mattia Pasini and Marco Simoncelli, who crashed during the session and restarted a little later.

Hector Faubel, Alex Debon and Ratthapark Wilairot close the top ten, while Alex Baldolini is 16th followed by Raffaele De Rosa, 17th and last Italian in the 250 second free practice rankings.
